Abstract
Οι πολιοϊοί, ο αιτιολογικός παράγοντας της πολιομυελίτιδας, είναι RNA ιοί θετικής
πολικότητας που ανήκουν στην οικογένεια των Picorna ιών και στο γένος των εντεροϊών. Το
μικρό μονόκλωνο RNA τους, μήκους 7500 νουκλεοτιδίων, περιβάλλεται από ένα μη
ελυτροφόρο καψίδιο εικοσαεδρικής συμμετρίας, με διάμετρο 28nm. Διακρίνονται σε τρεις
οροτύπους (PV1, PV2, PV3). Το πρόγραμμα εμβολιασμού κατά της πολιομυελίτιδας ξεκίνησε
στις αρχές της δεκαετίας του 1960 και με τη χρήση δύο εμβολίων, του IPV και του OPV,
επιτεύχθηκε η σχεδόν πλήρης εκρίζωση της ασθένειας στις ανεπτυγμένες χώρες. Τα
εξασθενημένα στελέχη Sabin του OPV εμβολίου, πολλαπλασιάζονται στην ανθρώπινη εντερική
κοιλότητα και σε σπάνιες περιπτώσεις μπορεί να προκαλέσουν εμβολιοσυνδεόμενη
παραλυτική πολιομυελίτιδα (vaccine-associated paralytic poliomyelitis - VAPP). Η γενετική
αστάθεια των Sabin στελεχών, η οποία έχει σαν αποτέλεσμα τη συσσώρευση μεταλλάξεων και
ανασυνδυασμών, αποτελεί μια από τις κύριες αιτίες εμφάνισης VAPP. Τα ιικά στελέχη που
απομονώνονται σε τέτοιες περιπτώσεις ονομάζονται VDPVs (Vaccine Derived Polioviruses).
Στην παρούσα εργασία, μελετήθηκαν φαινοτυπικά χαρακτηριστικά, όπως η
θερμοευαισθησία και η κινητική ανάπτυξης, οχτώ OPV παραγόμενων στελεχών (έξι από τα
οποία ήταν ανασυνδυασμένα) και έγινε προσπάθεια συσχέτισής τους με γενωμικές
τροποποιήσεις (μεταλλάξεις και ανασυνδυασμούς). Όλα τα OPV-παραγόμενα στελέχη
εμφάνισαν μεταλλάξεις σε συγκεκριμένες θέσεις του ιϊκού γενώματος, οι οποίες σχετίζονται με
τον εξασθενημένο και θερμοευαίσθητο φαινότυπο των Sabin στελεχών. Τα αποτελέσματα
έδειξαν ότι τόσο οι μεταλλάξεις όσο και οι ανασυνδυασμοί θα μπορούσαν να επηρεάσουν τα
φαινοτυπικά χαρακτηριστικά των Sabin παραγόμενων στελεχών και ίσως να οδηγήσουν στην
αναστροφή των εμβολιακών στελεχών προς νευρομολυσματικά.
