Ανίχνευση μεταλλάξεων σε μιτοχονδριακά tRNA γονίδια από ασθενείς με ποικίλα κλινικά συμπτώματα

Abstract
Τα τελευταία χρόνια σύνδρομα με βαριά κλινικά συμπτώματα όπως το MELAS και
το MERRF έχουν συνδεθεί με την ανίχνευση μεταλλάξεων σε συγκεκριμένα
μιτοχονδριακά γονίδια που κωδικοποιούν μόρια tRNA. Το γονίδιο το οποίο
συγκεντρώνει τις περισσότερες σε αριθμό αναφερόμενες μεταλλάξεις, είναι αυτό του
tRNALeu(UUR) (MT-TL1). Δεδομένης της σημασίας των μορίων tRNA στη
μιτοχονδριακή πρωτεϊνοσύνθεση μελετήθηκε σε μία ομάδα 32 ασθενών με ποικίλα
κλινικά συμπτώματα η ύπαρξη μεταλλάξεων στην ευρύτερη περιοχή του MT-TL1
συμπεριλαμβανομένων και τμημάτων των παρακείμενων γονιδίων, MT-RNR2 (16S
rRNA) και MT-ND1 (αφυδρογονάση του NADH). Παράλληλα, η μελέτη
επεκτάθηκε και σε τρία επιπλέον μιτοχονδριακά tRNA γονίδια [tRNAIIls,
tRNASer(AGY) και tRNALeu(CUN)J. Ο αρχικός έλεγχος για την ύπαρξη ή όχι
διαφορετικού προτύπου στην ομάδα των ασθενών έγινε με SSCP και τα PCR
προϊόντα στα οποία εντοπίστηκαν διαφορές σε σχέση με τους μάρτυρες
αλληλουχήθηκαν προκειμένου να ταυτοποιηθεί η ακριβής θέση των μεταλλάξεων.
Καμία μετάλλαξη δεν ανιχνεύθηκε στο γονίδιο του tRNALeu(UUR). Παρατηρήθηκαν
μεταλλάξεις σε θέσεις που έχουν προαναφερθεί στα γονίδια tRNALeu(CUN); ND1 και
16S rRNA και οι οποίες έχουν συνδεθεί με συγκεκριμένα σύνδρομα σε ανθρώπινους
πληθυσμούς. Επιπλέον όμως βρέθηκαν δύο νέες μεταλλάξεις στα γονίδια των ND1
και 16S rRNA που δεν έχουν αναφερθεί προηγουμένως στη διεθνή βιβλιογραφία και
οι οποίες πιθανότατα σχετίζονται με τη γενικότερη κλινική εικόνα των ασθενών.
